Slide was placed on microscope stage. A single Nikon 10x BD Plan objective with outer sleeve removed mounted in turret via RMS to BD adapter. All other objective openings sealed with electrical tape. Microscope head has been replaced with extension tubes connected to a Canon t3i (600D) camera. Linear polarizer placed inside extension tubes. Mirror lockup, 2nd curtain shutter and remote control cable used to minimize vibration. Specimen illuminated with 4 LEDs with simple paper cylinder diffusion. 2790 separate 1 second exposures focus stacked and merged into a single 1.09 gigapixel image of the left dorsal view. Image duplicated and flipped horizontally to make a 2.19 gigapixel image. Contrast, color, and sharpening adjustments applied.
